RESUMO

The authors suggest a new improved automated method for separate measurements of urinary porphyrins (coproporphyrins and uroporphyrins) in native urine with computer processing of spectral data. The method of dividing the total spectrum of porphyrin fluorescence takes account of background non-porphyrin admixtures in the urine and corrects the data on the basis of regression analysis. The data of this method were compared to those of other widely used methods and were found to be in good correlation; the advantages of the automated method are discussed.

RESUMO

Content of coproporphyrin was estimated in urine of females with genital, thyroid and gastrointestinal cancer. A rate of coproporphyrin elimination with urine depended on the type of tumor and its site. Concentration of coproporphyrin was decreased in urine of patients with both malignant and benign tumors as compared with that of healthy persons.

RESUMO

Several parameters of chlorin e6 and its derivative chlorin e6 ethylenediamide have been investigated as these compound are potential sensitizers for photodynamic therapy. A study carried out to compare the cellular uptake of the pigments indicates that chlorin e6 ethylenediamide possesses an enhanced affinity for tumour cells and cellular membranes. Comparison of the uptake in induced sarcoma shows that chlorin e6 ethylenediamide is a much better tumour localizer than chlorin e6. The efficiency of phototherapy with chlorin e6 ethylenediamide is higher than that with chlorin e6. These data show the influence of the substitution of the carboxyl groups in chlorin e6 by ester and amide groups on the photosensitizing properties of the pigments.

RESUMO

Results of the analysis of a number of parameters which determine the efficiency of using the photodynamic action for treating rheumatoid arthritis are reported. The investigations are based on determining the character of sensitizer stabilization in the joint tissue and evaluating its stability. As the sensitizer chlorin e6 was chosen. We have established the fact of contrast accumulation of chlorin e6 in the synovial membrane and cartilage, developed the system of intrajoint introduction of the pigment, studied the kinetics of sensitizer destruction under irradiation.

RESUMO

Data on porphyrin metabolism disturbances under the action of ionizing radiation on the human body are presented. The studies are based on estimating the quantitative content of coproporphyrin in the diurnal urine diuresis of persons irradiated as a result of the Chernobyl accident. The results of examining the population of a number of the zones of close control in Mogilev and Gomel Provinces are considered. Unlike the presumably healthy subjects, many people of this group show a significant decrease of porphyrin excretion with urine. The discovered sensitivity to the action of radiation is individual. A preliminary estimation of the porphyrin test depending on the degree of morphological changes in the gastric mucosa was made during an all-round clinical examination of the irradiated subjects.

RESUMO

Levels of copro- and protoporphyrin in red blood cells obtained from patients with tumors of the stomach, large bowel and thyroid are discussed. The levels of both porphyrins proved markedly increased in patients with gastric and intestinal cancer but were nearly normal in cases of benign tumor of various sites and thyroid cancer.

RESUMO

Methods for urinary coproporphyrin measurements are analyzed. Data obtained by two methods, spectrophotometry and fluorescent method, are presented, the advantages and shortcomings of both are discussed. The authors recommend the fluorescent technique as a universal rapid method for wide medical practice. The problem of the units of coproporphyrin measurement is also discussed. The authors suggest replacing measurements per g of creatinine or daily portion by estimation of the coproporphyrin index in the morning portion of the urine. Study of the effects of various factors (climatic conditions, nutrition, patient's age and sex) on normal coproporphyrin excretion from the body has demonstrated that these factors do not noticeably influence porphyrin metabolism; for normal subjects the coproporphyrin index varies from 30 to 110 micrograms/day.

RESUMO

Content of copro- and protoporphyrins was studied in erythrocytes of women with tumor of genitals. Distinct increase in content of these porphyrins, especially of protoporphyrin, was detected in erythrocytes of the patients with malignant tumors as compared with the patterns of healthy persons.

RESUMO

Transition of the products of the first restoration of porphyrines to the second one in the darkness and practically in the absence of a reducer and oxygen has been studied. Its cause is the disproportioning of dyhydroform into tetrahydro- and the initial pigment. Regeneration of porphyrines under these conditions is also shown. The rate of both reactions significantly increases in the light.
